Antitumoral Action of Resveratrol Through Adenosinergic Signaling in C6 Glioma Cells

چکیده

Gliomas are the most common and aggressive primary tumors in central nervous system. The nucleoside adenosine is considered to be one major constituent within tumor microenvironment. level mainly depends on two enzymatic activities: 5′-nucleotidase (5′NT or CD73) that synthesizes from AMP, deaminase (ADA) converts into inosine. Adenosine activates specific G-protein coupled receptors named A 1 , 2A 2B 3 receptors. Resveratrol, a natural polyphenol present grapes, peanuts, berries, shows several healthy effects, including protection against cardiovascular, endocrine, neurodegenerative diseases cancer. However, molecular mechanisms of resveratrol actions not well known. Recently, we demonstrated acts as an agonist for rat C6 glioma cells. work aimed investigate involvement metabolism underlying antitumoral action resveratrol. Results presented herein show was able decrease cell numbers viability reduce CD73 ADA activities, leading increase extracellular levels. Some effects were reduced by blockade DPCPX MRS1220, respectively. These results suggest activity located plasma membrane addition fine-tuned modulatory role could involved, at least part, antiproliferative

Distinct Mechanisms Underlying Resveratrol-Mediated Protection from Types of Cellular Stress in C6 Glioma Cells

The polyphenolic phytostilbene, trans-resveratrol, is found in high amounts in several types and tissues of plants, including grapes, and has been proposed to have beneficial effects in the central nervous system due to its activity as an antioxidant. The objective of the present study was to identify the mechanisms underlying the protective effects of resveratrol under conditions of oxidative ...

Resveratrol and quercetin cooperate to induce senescence-like growth arrest in C6 rat glioma cells.

Glioma is the most frequent and malignant primary human brain tumor with dismal prognosis despite multimodal therapy. Resveratrol and quercetin, two structurally related and naturally occurring polyphenols, are proposed to have anticancer effects. We report here that resveratrol and quercetin decreased the cell number in four glioma cell lines but not in rat astrocytes. Low doses of resveratrol...

Prostaglandin E2 Synthesis in C6 Glioma Cells

Prostaglandin E2 (PGE2) plays an important role in immunosuppression and tumor growth. PGE2 inhibitors such as aspirin and indomethacin suppress experimental tumor growth. Little is known of the relationship between PGE2 synthesis in brain tumors and the dose of aspirin or indomethacin. The present study was undertaken to evaluate the effect of different doses of aspirin and indomethacin on PGE...

The Effects of Progesterone on Glial Cell Line-derived Neurotrophic Factor Secretion from C6 Glioma Cells

Objective(s)Progesterone is a steroid hormone whose biology has been greatly studied within the confines of reproductive function. In recent years, the neuroprotective effects of progesterone have attracted growing interest. Glial cell line-derived neurotrophic factor (GDNF), is a neurotrophic factor which plays a crucial role in the development and maintenance of distinct sets of central and p...

Dual action of chronic ethanol treatment on LPS-induced response in C6 glioma cells

In this study we investigated the anti-inflammatory effects of chronic ethanol (EtOH) treatment on lipopolysaccharide (LPS)-stimulated C6 glioma cells. The cells were chronically treated with 200mM EtOH; coincubation with LPS and EtOH was obtained upon addition of 2μg/ml LPS to the incubation medium in the last 24h of EtOH exposure. We found that EtOH prevented the LPS-induced production of tum...
